How do you know that the female leads in his book were always (if ever) based
on his wife?

Here, here. I was introduced rather late to the writings of the great man.
And I've always identified with his outlook. I can imagine that his 2nd
wife,
Jeanie, loved being immortalized forever as the female lead in his books,
too. The heroine was always her...and always identifible because of that.
They must have had quite a relationship.
